Details for Assay 9: RTgill-W1 cell line assay (fish cell line acute toxicity, OECD TG 249)
Description
Fish cells (permanent cell line from rainbow trout gill, RTgill-W1), cultured in a 24-well plate format, are exposed to a chemical for 24 h, followed by assessment of cell viability based on three fluorescent cell viability indicator dyes, measured on the same set of cells.Validation Notes
The assay was validated in a round-robin test prior to its adoption as the OECD Test Guideline No 249No biological action information submitted

| Fluorescent indicator dyes (indicative of cell viability / cytotoxicity), namely: a resazurin-based dye for measuring cell metabolic activity (e.g. AlamarBlue); 5-carboxyfluorescein diacetate acetoxy methyl ester (CFDA-AM) dye for assessing cell membrane integrity; and Neutral Red, which measures lysosomal membrane integrity. |
