Details for Assay 14: Aromatase activity by tritiated water release.
Description
Tritiated androstenedione, labeled in the 1beta position, is incubated with NADPH in the presence of microsomes or post-mitochondrial supernatant prepared from harvested tissues or cells. Aromatase present in the sample converts the labeled androstenedione to estrone, with the release of tritiated water in a 1:1 molar ratio with the amount of estrone product generated. Tritiated water is subsequently extracted by liquid-liquid extraction and quantified by liquid scintillation counting to determine the enzyme activity expressed in molar concentration of product produced per min per mg protein in the sample.Validation notes not yet submitted
