Details for Assay 14: Aromatase activity by tritiated water release.

Description
Tritiated androstenedione, labeled in the 1beta position, is incubated with NADPH in the presence of microsomes or post-mitochondrial supernatant prepared from harvested tissues or cells. Aromatase present in the sample converts the labeled androstenedione to estrone, with the release of tritiated water in a 1:1 molar ratio with the amount of estrone product generated. Tritiated water is subsequently extracted by liquid-liquid extraction and quantified by liquid scintillation counting to determine the enzyme activity expressed in molar concentration of product produced per min per mg protein in the sample.
